目的

研究过氧化物酶增殖物激活受体γ(PPARγ)对肝癌细胞迁移的影响,并探讨其机制。

方法

应用伤口愈合实验观察PPARγ的合成配体曲格列酮和罗格列酮对肝癌细胞系BEL-7402迁移的影响;应用免疫荧光化学、RT-PCR和Western Blotting分析探讨其中的机制。

结果

曲格列酮和罗格列酮均能抑制肝癌细胞的迁移活动,其原因是由于该两种药物激活PPARγ通路且具有配体依赖性。

结论

PPARγ活化能够抑制肝癌细胞的迁移。

Objective

To investigate the effect of peroxisome proliferators-activated receptorγ(PPARγ)on migration of hepatocellular carcinoma cells and its underlying mechanisms.

Methods

Migration of HCC cell line BEL-7402 with the treatment of troglitazone and rosiglitazone was measured by a wound healing assay.PPARγmRNA and protein were assessed by RT-PCR,immunofluorescence chemistry,and Western Blotting analysis,respectively.

Results

Troglitazone and rosiglitazone inhibited the migration of BEL-7402 cells,both medcines activated PPARγsignaling pathway on ligand-dependent effect.

Conclusion

PPARγactivation inhibited the migration of HCC.
