目的

探讨E-选择素在肝癌特异性转移中的作用,并筛选有临床应用前景的抗肝癌转移药物。

方法

检测E-选择素在肝静脉内皮细胞(ED25)中的表达,检测活化ED25与肝癌细胞HepG2、结肠癌细胞Love、鼻咽癌细胞在体外的黏附差异,并测试不同药物对黏附的影响。

结果

E-选择素高强度表达于活化后的ED25,且能介导HepG2与ED25的黏附。HepG2条件培养液处理ED25可增强与HepG2、Love、鼻咽癌细胞的黏附,但HepG2和Love与ED25的黏附明显强于鼻咽癌细胞(P<0.05),而HepG2和Love与ED25的黏附差异无显著性(P=0.29);抗E-选择素抗体、地塞米松、氧化亚砷、吡咯烷二硫氨基甲酸盐(PDTC)有较强抑制肝癌细胞与ED25黏附作用。

结论

E-选择素可促进肝癌细胞与ED25的黏附;肝癌细胞可分泌促进ED25表达E-选择素表达的物质,从而促进了肝癌的转移;地塞米松、氧化亚砷、PDTC可以抑制肝癌细胞与ED25的黏附,有望成为预防肝癌转移药物。

Objective

To explore the role of E-selectin on the metastasis of hepatocelluar carcinoma.To select related possible drugs to inhibit transferance of hepatocelluar carcinoma.

Methods

Expression of E-selectin in hepatic vein endothelial cell (ED25) was detected. Difference of adhesion between ED25 and HepG2, colon carcinoma, nasopharyngeal carcinoma cells were compared in vitro by solid phase adhesion assay.A number of drugs were tested for the affects on the tumor cell-endothelial adhesion.

Results

E-selectin was high expressed on the surface of actived ED25 and could mediate the adhesion between HepG2 and ED25. The adhesion were enhanced after ED25 was stimulated by HepG2 condition medium in different tumor cells. Anti-E-selectin antibody,dexamethasone,arenous oxide and PDTC were able to block the adhesion between HepG2 to ED25 at low concentration.

Conclusion

E-selectin is a important molecular in the adhesion of hepatoma cells to ED25. HepG2 can secrete some factors to promote the expression of E-selectin. Agents capable of inhibiting NF-κB can be hopefully used as anti-adhesion drugs to inhibit metastasis of hepatocelluar carcinoma.
