目的

明确成年鼠胰腺中有无胰腺干细胞存在,了解其可能的分化机制。

方法

切除大鼠大部分胰腺(>90%)并消化、筛选出胰腺干细胞后回植入大鼠自体皮下,予不同处理因素诱导。观察大鼠血糖、血胰岛素浓度以及糖耐量试验的变化,并取移植物作常规染色和胰岛素、角蛋白19(CK-19)抗原的免疫组化检测。

结果

植入自体胰腺干细胞8周起,受体鼠血糖水平(P<0.01)及血胰岛素水平(P<0.05)与对照组比较均有明显差异;而对突发的糖刺激的耐受能力A组高于对照组,P<0.05,B组则在给糖后180 min表现出与对照组的差异,P<0.05;术后10~12周受体小鼠移植物中可见与大鼠腹腔胰腺组织近似的乳白色内含细小颗粒结构,以及胰岛素染色及CK-19染色阳性细胞,而对照组均为发现类似结构及细胞。

结论

成体大鼠胰腺非内分泌组织中存在胰腺干细胞,经移植后能分化为胰岛素分泌细胞,并有改善受体鼠高血糖状态和糖耐量异常的作用。

Objective

To make sure whether there are stem cells in the pancreas of adult rats and to discover its possible mechanisms of differentiation.

Methods

Most of the pancreas in rats (>90%) were resected and digested, then screened out pancreatic stem cells.We implanted these cells in rats subcutaneously, and induced them with different treatment factors.The changes of blood sugar,insulin and glucose tolerance of the rats, immunohistochemistry testing of insulin, CK-19 antigen and HE staining were observed from the grafts.

Results

After islet stem cell transplantation, blood sugar of recipient groups were lower than control groups, while blood insulin and the ability of reaction to sudden stimulation of sugar were higher than control groups, insulin positive cells also seen in the grafts.

Conclusion

There are stem cells in the non-endocrine tissue of pancreas of adult rats, which can differentiate into insulin-secrete cells and show ability to improve blood sugar, insulin level and glucose tolerance condition in the recipient rats after transplantation.
