目的

探讨巨噬细胞对SW480结肠癌细胞侵袭和迁移的影响及可能机制。

方法

流式细胞仪检测共培养体系中不同激活状态的巨噬细胞对SW480细胞凋亡和细胞周期的影响,侵袭实验、划痕实验检测巨噬细胞对SW480结肠癌细胞侵袭和迁移的影响,酶联免疫吸附(ELISA)方法检测Transwell共培养体系上清中血管内皮生长因子(VEGF)的变化。

结果

空白对照组、非激活组、IL-4激活组SW480细胞凋亡及S期细胞百分数差异无统计学意义,IL-4激活的巨噬细胞明显抑制SW480细胞的侵袭和迁移(P<0.05)。激活组的VEGF浓度显著降低(P<0.05)。

结论

IL-4激活的巨噬细胞能够抑制SW480结肠癌细胞的侵袭和迁移,其机制可能与下调VEGF的分泌有关。

Objective

To observe the influence of IL-4-activated macrophage on the invasion and migration of SW480 colon cancer cell, and investigate its possible mechanism.

Methods

Macrophages were activated by IL-4 and co-cultured with SW480 colon cancer cell. Flow cytometry was used to detect the apoptosis and cell cycle. Boyden chamber assay and wound healing assay were used to detect the migration and invasion of the SW480 colon cancer cell under the effects of differently activated macrophages. The concentration of vascular endothelial growth factor (VEGF) of the co-cultured supernatant was detected by enzyme linked immunosorbent assay(ELISA).

Results

There were no significant differences of the percentages of apoptosis and S phase cancer cells in all groups. Boyden chamber assay and wound healing assays showed that the migration and invasion of cancer cells were significantly inhibited in group of co-cultured with the IL-4-activated macrophage than in other groups, P<0.05. The concentration of VEGF of the supernatant in IL-4-activated macrophage group was much lower than other groups, P<0.05.

Conclusion

IL-4-activated Macrophage inhibits the invasion and migration of SW480 colon cancer cell. The mechanism may be correlated with inhibiting VEGF secretion of SW480 cell.
