目的

探讨糖酵解抑制剂2-脱氧-D-葡萄糖(2-DG)与氟尿嘧啶(5-Fu)单独及联合应用对人胃癌细胞MGC-803增殖及凋亡的影响。

方法

实验设立2-DG组、5-Fu组、联合组(2-DG +5-Fu)及空白对照组,将2-DG(10 mmol/L)及5-Fu(5.0 μg/L)单药及联合用药作用于人胃癌细胞MGC-803,MTT法检测各组MGC-803细胞的增殖抑制率,流式细胞仪检测各组细胞凋亡情况。

结果

2-DG和5-Fu均可使胃癌细胞增殖受到抑制并可诱导细胞凋亡,两药合用后其作用明显增强。

结论

2-DG能有效抑制人胃癌细胞MGC-803细胞生长增殖,并诱导其凋亡;2-DG与5-Fu联合应用抗肿瘤作用明显增强。

Objective

To investigate the effects of 2-deoxy-D-glucose ( 2-DG) or 5-Fu alone and the combination of both on the proliferation and apoptosis of human gastric cancer MGC-803 cells.

Methods

Human gastric cancer MGC-803 cells were treated with 2-DG or 5-Fu alone, or both. The inhibitory effect on the proliferation of MGC-803 cells was detected by MTT method. Apoptotic rate was assayed by flow cytometry.

Results

2-DG or 5-Fu inhibited the growth of MGC-803 cells obviously and induced the apoptosis. When 2-DG and 5-Fu were combined, the inhibitory effect and apoptotic rate were improved (P<0.05).

Conclusions

2-DG inhibits the growth of human gastric cancer MGC-803 cells. The combination of 2-DG and 5-Fu improves the ability of 5-Fu to attack the tumor cells.
