目的

探讨15-脱氧-前列腺素J2(15-d-PGJ2)对肝癌细胞SMMC-7721侵袭转移的影响及其内在的机制。

方法

SMMC-7721肝癌细胞经天然配体15-d-PGJ2和(或)抑制剂GW9662处理,用伤口愈合实验和Transwell方法检测细胞的转移侵袭能力,用RT-PCR检测细胞PTEN和MMP-9 mRNA的表达,用Western blotting检测细胞PTEN和MMP-9蛋白的表达。

结果

15-d-PGJ2对SMMC-7721细胞的转移侵袭具有抑制作用,并能上调PTEN mRNA和蛋白的表达,下调MMP-9 mRNA和蛋白的表达;单药GW9662无此效应,GW9662与15-d-PGJ2联合能够逆转此效应。

结论

15-d-PGJ2是通过激活PPAR信号上调PTEN表达,下调MMP-9表达,抑制肝癌细胞的侵袭转移。

Objective

To investigate the effect of 15-deoxy-prostaglandin J2 (15-d-PGJ2) on the invasion and metastasis of hepatocellular carcinoma SMMC-7721 cells, as well as its underlying mechanisms.

Methods

Hepatocellular carcinoma SMMC-7721 cells were treated by natural ligand 15-d-PGJ2 with or without antagonist GW9662 of peroxisome proliferator-activated receptor (PPAR). Invasion and metastasis of SMMC-7721 cells was measured by wound healing assay and Transwell chambers. mRNAs of PTEN and MMP-9 were assessed by RT-PCR and proteins by Western Blotting analysis, respectively.

Results

15-d-PGJ2, but not GW9662, inhibited the invasion and metastasis of SMMC-7721 cells, up-regulated PTEN mRNA and protein expression, and down-regulated MMP-9 mRNA and protein expression. However, after SMMC-7721 cells were pretreated for 1 h with GW9662 and exposed to 15-d-PGJ2, the inhibitive effects of 15-d-PGJ2 on invasion and metastasis of cancer cells were reversed.

Conclusion

15-d-PGJ2 increases PTEN expression and decreases MMP-9 expression to suppress the invasion and metastasis of HCC via activating PPAR signal pathway.
