Expression and significance of transcription factor Gli2 of Hedgehog signaling pathway in human hepatocellular carcinoma

Abstract:

Objective

To investigate the expression and clinical significance of downstream transcription factor glioma-associated oncogene homologue 2(Gli2) of Hedgehog(Hh) signaling pathway in human hepatocellular carcinoma(HCC).

Methods

Real time RT-PCR and Western blot were used for detection of mRNA and protein of Gli2 in 20 pairs of HCC tissues and corresponding paraneoplastic liver tissues; Immunohistochemistry was used for detection of Gli2 protein in 52 pairs of HCC tissues and paraneoplastic liver tissues.

Results

mRNA and protein expression of Gli2 in HCC were significantly higher than in paraneoplastic liver tissues. Immunohistochemical results showed that positive expression of Gli2 protein was 38/52(73.1%) in HCC tissues and 8/52(15.4%) in corresponding adjacent tissues, the difference was significant(P<0.01) and the expression of Gli2 protein was correlated with tumor differentiation, vascular invasion and lymph node metastasis(P<0.05).

Conclusion

The abnormal expression of Gli2 is closely related to tumorigenesis, invasion and metastasis of HCC, Gli2 may be an important biological marker of HCC and biological therapeutic target.
