Experimental research of treatment of diabetes using islets from adult pancreata

Abstract:

Objective

To investigate the efficiency of adult pancreatic islets in the treatment of diabetic patients.

Methods

Thirteen adult donor panceata were used for this study. After collagenase digestion, the islets were isolated and purified followed by transplantation into diabetes SPF nude mice. Control group were transplanted by normal saline solution. Islet graft survival was monitored by the detection of glucose changes in diabetic mice and histological examination.

Results

Islet yield was (3608±403) IEQ /g in this study, while that was (2820±318)IEQ/g after purification with a purity of 85%. The blood glucose of 84.6% diabetic mice decreased to normal in the first day post-transplantation. Normal blood glucose level maintained for a long time. There was significant difference in the Kaplan-Meier analysis for the islet survival (P = 0.0012).

Conclusion

The adult donor pancreas is an ideal source for islet transplantation due to high yield and good function with a high purity and viability to correct the diabetic nude mice blood glucose level.
