Effect of interleukin-2 gene transfection on pulmonary metastasis of breast cancer in nude mice

Abstract:

Objective

To evaluate the effect of interleukin-2 (IL-2) gene transfection on treating pulmonary metastasis of breast cancer in nude mice.

Methods

Breast cancer lines MD231 and BT474 were first transfected with pORF-hIL-2 plasmid, then cell invasiveness and migration capability were tested. Thirty nude mice with pulmonary metastasis of breast cancer were randomly divided into pORF-hIL-2 plasmid transfection group and the control group, then the mixture of liposome and pORF-hIL-2 plasmid was injected into their tumors subcutaneously, and finally the volumetric change of the tumors and the lung metastic lesions was observed.

Results

The cell invasiveness and migration capability diminished in MD231 and BT474 after transfected with pORF-hIL-2 plasmid. Compared with the control group, the volumetric growth of the subcutaneous nodular tumors in the experiment group were inhibited obviously, and the difference was statistically significant (t=-8.214, P<0.001); after the study period, the difference of the number of the lung metastic lesions in the two groups were statistically significant (t=3.854, P=0.007).

Conclusion

IL-2 gene plasmid intratumoral injection has shown to improve anti-tumor effects significantly to treat pulmonary metastasis of breast cancer in nude mice, and to inhibit lung metastatic lesions effectively.
