Expression of circadian rhythm gene Timeless in different parts of rectal cancer and its relationship with clinical characteristics and prognosis

Abstract:

Objective

To analyze the expression of biorhythmic gene Timeless in different parts of rectal cancer and their relationship with prognosis, so as to provide more possibilities for individualized treatment and prognosis monitoring.

Methods

From April 2012 to April 2015, 384 patients with rectal cancer admitted to Branch Courts of Huaian First Peoples Hospital were selected as source of specimens. According to the location of the tumors, they were divided into high group (n=64), middle group (n=122) and low group (n=198). The Real-time fluorescence quantitative PCR and immunohistochemistry were used to detect the expression of Timeless mRNA and protein in cancer tissues and adjacent normal tissues, and to analyze their relationship with clinical characteristics and survival.

Results

Timeless mRNA and protein in cancer tissues of high, middle and low rectal group were higher than those in normal adjacent tissues (P<0.01). The difference of Timeless mRNA and protein among the three groups was statistically significant (P<0.05), which was low group > middle group > high group. There were differences in tumor size, differentiation, TNM stage and survival in Timeless mRNA and protein. Cox regression analysis showed that tumor location, TNM stage and Timeless were independent risk factors for prognosis of rectal cancer (P<0.05).

Conclusion

Timeless is highly expressed in rectal cancer, especially in low rectal cancer, which is closely related to tumor size, differentiation, TNM stage and survival, and is one of the independent risk factors affecting its prognosis.
