Study of proliferation inhibition of octreotide on colonic cancer cells

Abstract:

Objective

To study the effect of somatosatin analogue octreotide at different concentrations on proliferation in colonic cancer cell line SW480.

Methods

To cultivate colon cancer SW480 cell in vitro, MTT assay was used to analyze the effect of octreotide at different concentrations on the proliferation of SW480 cell. The best inhibiting concentration was screened which had the maximum potency. Colony formation assay on plate was used to test inhibition of clone formation in SW480 cells by octreotide(OCT).

Results

Different concentrations of OCT inhibited the proliferation of SW480 cells, in dose and time-dependent manner during the concentrations of OCT(1×10-10 to 1×10-8 mol/L), and the inhibitory effect of octreotide at 1×10-8 mol/L was the most significant. Colony formation assay on plate showed that octreotide significantly inhibited the anchorage dependent growth of SW480 cells in a concentration-dependent manner.

Conclusion

OCT causes inhibition of colonic cancer cell line SW480 growth and proliferation, and the effect was dose and time dependent to a certain degree.
