Expressions of lncRNA GHET1 and IGF2BP1 in colorectal cancer and their influence on prognosis

Abstract:

Objective

To investigate the expressions of long non-coding RNA gastric carcinoma highly expressed transcript 1 (lncRNA GHET1) and insulin-like growth factor mRNA-binding protein 1 (IGF2BP1) in colorectal cancer (CRC), and to explore their relationship with the prognosis of CRC.

Methods

From January 2014 to January 2015, tissue samples from 90 CRC patients with complete pathological and clinical data in South Hospital of the Sixth People’s Hospital Affiliated to Shanghai Jiaotong University were selected as the study group, and the adjacent tissue samples were taken as the control group. The expressions of GHET1 and IGF2BP1 were detected by real-time fluorescence quantitative PCR (qPCR) and immunohistochemistry. The relationship between the expressions of GHET1 and IGF2BP1 and clinicopathological features was analyzed. Kaplan-Meier method was used to analyze the relationship between the expressions of GHET1 and IGF2BP1 in CRC tissues and prognosis, while Cox regression analysis was used for the prognostic factors of CRC.

Results

The level of GHET1 mRNA and the positive expression rate of IGF2BP1 protein in the study group were significantly higher than those in the control group (P<0.05). The expressions of GHET1 mRNA and IGF2BP1 protein were correlated with tumor differentiation, depth of invasion, TNM stage and lymph node metastasis (P<0.05). Kaplan-Meier analysis showed that the survival rate of patients with high expression of GHET1 mRNA and IGF2BP1 protein positive was lower than patients with low expression of GHET1 and IGF2BP1 negative (32.80% vs 75.00%, χ2=17.775; 33.33% vs 80.00%, χ2=13.615; both P<0.05). Multivariate Cox analysis showed that high expression of GHET1 mRNA and positive expression of IGF2BP1 protein were risk factors for the prognosis of CRC (P<0.05).

Conclusion

There are high expressions of GHET1 mRNA and IGF2BP1 protein in CRC tissues, which are closely related to the poor prognosis of patients.
