Effect of RhoC gene on the proliferation and invasion abilities of human QBC939 cholangiocarcinoma cell

Abstract:

Objective

To explore the ras homolog gene C(RhoC)on the proliferation and invasion properties of cholangiocarcinoma cell line(QBC939).

Methods

The RhoC cDNA was transfected into human cholangiocarcinoma cell line(QBC939)with lipofectane.The cell growth curve was made to determine the inhibitory rate of cells, flow cytometry was used to analyze the changes of cell cycle of the tumor cells,boyden chamber was made to detect the invasive ability of cells before and after the gene transfection.

Results

RhoC cDNA could enhanced the growth of cholangiocarcinoma cell,and their invasiveness increased,compared with those before transfected. Flow cytometry showed lowerpercentageoftransfected cellsin G1 phasethan thatofuntransfected cells.

Conclusion

RhoC gene can enhance the abilities of proliferation and invasion in cholangiocarcinoma cell line in vitro.
