Study of LPA-induced VEGF secretion in gastric cancer

Abstract:

Objective

To examine the relation between lysophosphatidic acid (LPA)and vascular endothelial growth factor(VEGF)in cultured peritoneal mesothelial cells(PMC)in gastric cancer.

Methods

Human PMC stimulated with LPA were isolated and cultured in vitro.The secretion changes of VEGF in PMC were examined by ELISA.Then PMC was incubated with PTX or Ki16425,which were antagonists of LPA.Effect of PTX on LPA-induced VEGF secretion was determined.

Results

VEGF secretion in LPA-induced PMC increased in a dose-dependent manner.After PMC were treated with PTX or Ki16425,VEGF secretion were significantly reduced.

Conclusion

LPA-induced VEGF secretion may have an important role in the growth of cancer cells disseminated in the peritonealcavity.
