Abstract:

Objective

To investigate lymphangiogenesis in paritumor and intratumor tissue of gastric cancer by detecting the expression of specific lymphatic endothelium marker of lymphatic vessel hyaluronan receptor-1(LYVE-1) and vascular endothelial growth factor receptor-3(VEGFR-3).

Methods

LYVE-1,VEGFR-3 and their mRNA were examined in 62 gastric cancer tissue by routine immunohistochemical method and real time polymerase chain reaction respectively. Then the density of lymph vessel and the content of LYVE-1 and VEGFR-3 mRNA were calculated.

Results

The density of lymph vessel was higher in paritumor than intratumor tissue. The expression of LYVE-1 and VEGFR-3 protein could not be detected in normal tissue. The density of lymph vessel in paritumor tissue and the mRNA content of LYVE-1 and VEGFR-3 in cancer tissue were correlated with invasion depth, lymphnode metastasis, tumor stage and postoperational recurrence (P<0.05).

Conclusions

LYVE-1 is one kind of more specific lymphatic endothelium marker than VEGFR-3.Lymphangiogenesis can promote the progress and lymph node metastasis in gastric cancer.

Key words: Gastric neoplasm, Hyaluronan receptor, Vascular endothelial growth factor receptor-3, Polymerase chain reaction, Lymphangiogenesis
