Expression of Minichromosome maintenance 2 mRNA in colorectal adenoma and its clinical significance

Abstract:

Objective

To quantitatively compare the difference of minichromosome maintenance protein 2(MCM2) mRNA expression in colorectal adenomas with different clinicopatholigic characters. The aim was to further investigate the potential role of MCM2 in early diagnosis.

Methods

One hundred and twelve colorectal adenomas were collected, and each specimen was divided into two parts, one for immunohistochemistry and the other for real-time RT-PCR. Differences of MCM2 mRNA expression in colorectal adenomas with different clinicopatholigic characters were evaluated by REST-XL?.

Results

The expression of MCM2 mRNA was examined throughout epithelium in colorectal adenomas, and the expression of MCM2 mRNA in adenomas with high grade dysplasia was significantly higher than that in adenomas with low grade dysplasia(P = 0.032). MCM2 mRNA expression elevated in adenoma with villous type, larger size, sessile morphology and older age of patients, but the significant differences were not obtained by REST-XL?.

Conclusion

The expression of MCM2 in colorectal adenomas relative with grade dysplasia indicate the potential value of MCM2 in early diagnosis of colonic cancer.
