Influence of TGF-β1 on rat hepatic oval cell proliferation and apoptosis

Abstract:

Objective

To study the regulation of TGF-β1 on rat hepatic oval cells(HOCs)proliferation.

Methods

60 male Sprague-Dawley rats were randomly divided into the control group and the experimental group, and were established animal models of HOCs proliferation(2-AAF/PH). Using immunohistochemistry, RT-PCR and in situ detection of apoptosis, HOCs proliferation and dynamic expression of TGF-β1 of two model sets were observed at different time points.

Results

HOCs of the experimental group appeared on the second day after hepatectomy, and reached the peak on day 10. On day 16 oval cells significantly reduced and the structures of lobules of liver and hepatic cord recovered.TGF-β1 expression began to increase on day 6 after hepatectomy and reached its peak on day 16, and then it dropped to the normal level on day 21. TUNEL-positive cells in the experimental group reached its peak on day 16, in the late stage of the proliferation of oval cell's peak and in the same period of the expression of TGF -β1's peak.

Conclusion

TGF -β1 is closely related to HOCs proliferation and apoptosis, and it has negative regulated in HOCs proliferation.
