Abstract:
Objective
To investigate the preparation and the effect of T cell vaccine(TCV)in inducing immune tolerance of allogeneic in skin transplantation.
Methods
TCV made from Wistar rat recipients with the specific antigen of SD donor rats was used to immunize the rat recipients by peritoneal injection.Before and after the fifth day of the every times vaccination,one-way mixed lymphocyte reaction(MLR)were performed with effective cells of peripheral T cells of recipient rats and stimulator cells of peripheral T cells of SD rats,stimulator cells were attenuated by Mitomycin-C preliminary.To observed the effect of immune tolerance induction by TCV in rats,The skin grafts from SD rats were transplanted to Wistar rats.The grafts reactions and survival time were observed.Skin grafts histopathological examination were used as a reference to evaluate the reject reaction.
Results
The immunological responses were suppressed significantly after TCV vaccination(P﹤0.05).The surviving time and acute rejection of transplanted skins were better in TCV group(P﹤0.05).
Conclusion
In rat model,immunological tolerance can be successfully induced by TCV.TCV vaccination can increase the antirejection and surviving time of skin allografts.
