Abstract:

Objective

To determine the expression status of Shh and Ptch genes in hepatocellular carcinoma (HCC).

Methods

Tissue marray technique, immunohistochemistry (IHC)and in situ hybridization(ISH)were used to evaluate the expression of shh and Ptch genes in 100 cases with HCC along with 25 cases adjacent benign tissues and 5 cases normal liver tissues.

Results

The positive expression rates of Shh and Ptch in HCC detected with IHC were 19.28% (16/83)and 24.68%(19/77)respectively,the positive expression rates of both of these two genes in HCC were 44.29%(31/70)and 15.85%(13/82)when detected with ISH.The expressions of Shh and Ptch in HCC were elevated with the adjacent benign tissues and the normal liver tissues. The positive rates of Shh and Ptch genes in HCC were correlated with the tumor's size and pathological differentiation respectively.

Conclusions

The elevated expression of Shh and Ptch and the activation of Shh signaling pathway participate in the generation ofHCC.They may be ideal targets for therapy against HCC.
